C. elegans is renowned for the complete and largely invariant connectome, which has made a profound impact on understanding neural circuits and behavior. How the connectome arises through development is largely unknown. Combining live imaging, EM and network analysis, we previously found that the connectome can be partitioned into modules based on the strength of connections and the corresponding axons are stratified in the primary neuropil/brain of C. elegans called the nerve ring. Here, we show the developmental scheme that organizes the nerve ring structure. A chain of multicellular rosettes nucleates and partitions pioneer fascicles, and guides the fascicles to assemble into a pioneering scaffold with discrete entry points. Dynamic modeling of follower innervation showed that stereotypic axon positioning, stratification as well as the known bi-modal natural variation emerge without the commonly expected complex combinatorial adhesion code for axon-axon recognition. Expression of the CAM adhesion molecules in scRNA data is consistent with such a model. Our findings demonstrate how macro-level brain features emerge from collective cell behaviors even before neurite outgrowth.

鲍志戎教授任职于纽约纪念斯隆·凯特琳癌症中心,发育生物学方向。鲍教授在吉林大学获得学士学位,在圣路易斯华盛顿大学获得博士学位,并进行博士后研究。鲍教授深耕于体内单细胞分辨率下的基因网络和细胞行为。其团队结合显微镜技术、计算工具开发及遗传学方法,研究秀丽隐杆线虫胚胎发育中谱系分化及组织形态发生中的新型集体细胞行为调控机制,并通过广泛的合作将此系统扩展到果蝇、斑马鱼、小鼠和人类的研究,揭示保守基因网络和细胞行为机制。
