报告摘要:

Neurons grow cell-type specific dendrite arbors to receive and integrate information, which is critical for neural circuit formation. However, the molecular mechanisms which control dendrite arborization have not been fully elucidated. C. elegans PVD sensory neurons grow highly branched dendrites to innervate both skin and body-wall muscle cells. We have performed large-scale genetic screens to isolate mutants which are defective in PVD dendrite arborization. In my talk, I will introduce two recent studies in the lab. In the first part, we find CHDP-1, a cell cortex-localized protein, functions in the PVD neuron to promote cortical actin assembly, which is critical for proper dendritic development and transport of various types of organelles. Loss of chdp-1 causes defects in neuropeptide secretion and proprioception. Interestingly, these defects could be fully suppressed by further inactivating a protein kinase called SAX-1. In the second part, we find the C. elegans skin cells use an EAT-17-BICD-1-dynein-dependent endocytosis machinery to downregulate SAX-7/L1CAM, a dendrite branching ligand, to inhibit ectopic dendrite arborization in the lateral region of the worm body.
