报告摘要:
Plasma membrane, due to lipid-lipid and protein-lipid interactions, is segregated into functional membrane microdomains (FMMs) for various cellular activities. The formation of these microdomains is facilitated by scaffold proteins, as well as by spectrin-based membrane skeleton. Through cryo-EM structural analysis of the native spectrin-actin junctional complex from red blood cells, which is a highly decorated short filamentous actin (F-actin) acting as a central organizational unit of the erythrocytic membrane skeleton, we have revealed the general principles underlying the organization and assembly of the skeleton and elucidated specific molecular roles of the junctional complex components. More recently, from red blood cell derived vesicles, we discovered that ABCC4 forms four-started helical arrays on the membrane, resulting in the formation of tubular membranes. This result highlights an unrecognized, potential role of ABC-transporter in the shape maintenance of red blood cells. Altogether, these structural data manifest the potential of cryo-EM in the biophysical study of the structure and dynamics of biological membrane systems.

高宁实验室围绕着生命科学中的基础问题,致力于与重大疾病发生发展相关的蛋白质复合物的结构和机制研究。近年来在超大生物分子复合物的组装和调控机制等领域取得系列进展,例如血红细胞膜骨架系统的组装和结构基础、核糖体在细胞内组装的分子途径、组装因子的功能及机制等。
