报告摘要:
Genome editing technology has made significant advances in plant biotechnology in recent years, enabling the creation of genome-edited (GE) crops for biomass and food production. Engineered sequence-specific nucleases (SSNs), such as the clustered regular interval shortened palindrome repeat (CRISPR)/CRISPR-associated protein 9 (CRISPR/Cas9) system, which generate double strand breaks (DSBs) at target DNA sites, have laid the foundation for gene editing in plants. These generated DSBs are repaired mainly by the non-homologous end-joining (NHEJ) pathway and rarely by the homology-directed repair (HDR) pathway. The error-prone NHEJ pathway is the main repair mechanism for site-specific DSBs induced by SSNs, resulting in random mutations, whereas the HDR pathway produces error-free and precise knock-ins and sequence substitutions that modify the target sequence to the intended sequence, if an appropriate and specific repair donor template is provided. Therefore, in many organisms, HDR-mediated gene targeting (GT) is a powerful tool for precise genome editing. However, the extremely low efficiency of HDR has hindered GT in seed plants.
Recently, we have established a simple and highly efficient GT in Arabidopsis and rice via the CRISPR/Cas system. Using our GT technology, precise amino acid substitutions and knock-ins were achieved. Further analysis revealed the underlying molecular mechanism of GT, in which single-stranded DNA released from Agrobacterium during the transformation process likely serves as a donor template for the HDR.
