报告摘要:Abscisic acid (ABA) is a critical phytohormone involved in multifaceted processes in plant metabolism and growth under both stressed and nonstressed conditions. Its accumulation in various tissues and cells has long been established as a biomarker for plant stress responses. To better understand ABA distribution and dynamics at subcellular resolution, we have developed a series of soluble and nanobody-based membrane-anchored ABA probes that are targeted to different sub-organelles. Using these organellar sensors, it was revealed that organellar ABA in different cells responded distinctly depending on environmental stimuli. Besides, we identified ABCG16 as a dual transporter in ABA signaling for mediating both cellular ABA efflux and ER entry of ABA-GE. FRET (Förster resonance energy transfer) analysis on the conformational status of ABCG16 dimers further disclosed specific alterations in the conformation of ABCG16 homo- and heterodimers in response to ABA and ABA-GE, respectively. Together, we provide a mechanistic framework for understanding substrate specificities that are linked to different endomembrane during ABCG transporter-mediated ABA homeostasis.

个人简介:周叶玲,南方科技大学生命科学学院研究助理教授。2016年在荷兰乌特勒支大学取得博士学位后,回国加入南方科技大学,在该单位相继以博士后、研究助理教授等身份继续从事与植物激素相关的研究工作,尤其致力于细胞器ABA感应器的开发及应用研究。成功开发出一系列可溶性以及基于纳米抗体的膜锚定细胞器特异性ABA感应器,对植物亚细胞水平上的ABA浓度实现了可视化实时动态检测。目前的研究兴趣主要集中在转运蛋白如何在细胞不同内膜上实现特异性底物的转运机制。
